This practical course is intended to give you an opportunity to learn the basic techniques routinely used in biochemistry and molecular biology. The students will be introduced to the theory that serves the basis for these techniques.It will include discussion of the theoretical principles, practical details and applications of the key experimental techniques, including those that have led to the emergence of the new disciplines of genomics, proteomics and bioinformatics.

The students should be able to perform the biochemical experiments independently and interpret the laboratory results. By the end of the course students will be familiar with a number of common biochemical techniques: use of pipettes, balances and the spectrophotometer; making buffers, methods of bimolecular determination and assay of enzyme. Biochemistry laboratory provide a systematic way of performing biochemical calculations, qualitative and quantitative analysis of carbohydrates, preparation of buffers, spectrophotometer analysis of bimolecular and enzyme assay.

Research Activities

In Bioinformatics laboratory, each student is given a domain area of access and assigned a computer to work using softwares to compare nucleotide, aminoacid sequences and establish evolutionary relationship amongst various organisms. Department has undertaken research exploring possible gene markers for diagnosis of diseases such as leptospirosis, typhoid, dengue fever etc. Various bioinformatics tools are being used to understand the structure of commercially important proteins and their folding pattern.

Research Activities

In Bioprocess lab students learn the complexities of enzyme assays, growth profile of various microbes in small and large scale fermentation process and production profile of various bioproducts. Fermenters from New Brunswick, USA and Bioengineering, Switzerland are being used to grow microorganisms in large scale and the growth conditions could be optimized in such pilot scale fermentation operation. The laboratory was recently upgraded for continuous culturing (chemostat) of recombinant micorganism for the production of penicillin acyclase enzyme with the funding from AICTE, New Delhi.

The isolation and purification of bioproducts are analysed in Down Stream processing lab comprising of High Pressure Cell Disruptor (Constant Systems, U.K.), Sonicator (Sonics, USA), FPLC (GE, USA), HPLC (Shimadzu, Japan), Refrigerated super speed centrifuge (Sorvall, USA), Ultracentrifuge (Beckman, USA) and High speed centrifuge (Hitachi, Japan).

To develop skills of students by providing hands on training in some of the aspects of chemical engineering. This will be a pre-requisite for lab courses in Bioprocess. At the end of the course, the student would have learnt about fluid flow measurement like Venturimeter, Orificemeter etc., and fundamental concept of Heat Transfer and Mass Transfer background like Types of Heat Exchangers and Types of Distillation respectively and how to perform them. This will be very useful for specialized project and research work.
In this lab students are exposed to the chemical engineering instruments which have industrial applications. This lab focuses on the application of fluid mechanics, heat and mass transfer. Students practically understand the functioning of various flow meters, heat exchangers, flow through packings which have applications in the field of bioprocess industries and biopharma industries.

Gene Expression laboratory (GEL) laboratory has state of the art equipments to impart knowledge and skills in molecular biology, genetic engineering and immunology techniques. The students are being trained individually in performing each experiment with utmost care and purpose. GEL has an inbuilt mammalian cell culture facility where students are trained to handle several cell lines.
The infrastructure in GEL is utilized to train the students during their regular laboratory courses and research projects which are part of their B.Tech Biotechnology curriculum. Teaching in molecular biology labs are aimed at exposing students to DNA based techniques, bacterial cell transformation, electrophoresis and protein analysis. Experiments cbulletied out includes competent cell preparation, transformation by electroporation and heat shock method, plasmid and genomic DNA isolation, Polymerase Chain Reaction (PCR), agarose gel electrophoresis of DNA, restriction digestion of DNA and molecular weight calculation, SDS-PAGE electrophoresis to resolve proteins.
In the Genetic Engineering laboratory course, various DNA manipulation techniques and protein expression experiments are cbulletied out. Experiments such as large scale plasmid isolation, restriction analysis of plasmid vector constructs, designing primers to amplify target gene, PCR amplification of desired gene, plasmid vector manipulation, purification of restricted vector and PCR products, TOPO cloning, ligation and transformation, screening for recombinant clones and expression of recombinant proteins are performed. In the immunology lab, various immuno-chemistry techniques are taught. Animal handling, immunization and raising antisera, blood grouping, immuno-diffusion assays, immuno-electrophoresis, enzyme linked immuno-sorbent assay (ELISA), western blot, and antibody capture assays are cbulletied out.

Student research projects performed in the areas like cloning of viral coat protein gene, expression and purification of recombinant proteins, molecular characterization and phylogenetic analysis in microorganisms and plants, in-vitro studies on anti-cancer activities of homeopathy drugs. All the experiments cbulletied out in GEL come within the mandate of Institutional Biosafety Committee (IBSC) which is recognized by DBT, New Delhi.

Separation and purification of proteins and other compounds of interest from large scale fermentations or modified natural sources is a critical element of industrial biotechnology. It represents the major manufacturing cost, and therefore competitive advantage will depend on innovation and optimization of down stream processes.

Ongoing activities in the lab include purification of recombinant fusion- protein, modified biomaterials, intracellular enzymes involved in antibiotic synthesis and commercially important pigments.

Research Activities

Animal tissue culture lab is equipped with instruments such as CO2 incubators(Thermo, USA), Biosafety hoods and Fluorescent Microscope (Carl Zeiss, Germany) is actively functioning and involved in research establishing various mammalian cell lines, checking the expression profiles of protein and performing biological assays

Research Activities

Plant tissue culture lab where plants are grown under controlled condition is generating callus for the production of commercially important secondary metabolities. Development of transgenic plants, micropropagation of high value commercial crops, development of suspension cultures for the production of metabolites are being attempted. The major objective of these projects is to apply intensive technical knowledge to produce commercially important bioproducts in large scale. Antibiotic formulations and antimicrobial activities of herbal preparation are also being evaluated.
